Key instrumentation challenges in single-use media preparation
In single-use media preparation the biggest challenges are usually no the mixing bag itself, but the quality and reliability of the measurements use to verify media preparation. Media preparation remains one of the least-measured unit operations in single-use bioprocessing.
- Obtaining reliable pH measurements in disposable systems: Disposable pH sensors can exhibit drift, limited calibration capability and lower reliability compared with conventional reusable probes.
- Verifying complete powder dissolution and mixing homogeneity: Instrumentation often cannot directly confirm that all powder is dissolved and uniformly distributed.
- Accurate bag volume/level measurement: Flexible bags change shape during filling and emptying, making precise level measurement difficult.
- Maintaining data integrity with single-use sensors: Disposable instrumentation must still meet GMP data requirements while integrating into plant automation systems.
